Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 is a 49–55 kDa transcriptional regulator, defective in scurfy mice. High Foxp3 expression is observed in CD4+CD25+ regulatory T cells (Treg cells).

Immunoblotting with FJK-16s maps the epitope to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced, with both isoforms present in CD4+CD25+ lymphocytes. Mouse splenocytes lack this splice variant, suggesting species-specific regulation.

This antibody is optimized for use with the Foxp3 Transcription Factor Buffer Staining Set (Product #00-5523-00). Other buffers are not recommended.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated using Foxp3 buffers and mouse splenocytes. Recommended usage ≤0.06 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Titrate for optimal performance.

eFluor® 450 is an alternative to Pacific Blue®, emitting at 445 nm when excited at 405 nm (Violet laser). Ensure instrument compatibility.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is expressed in CD25+CD4+ regulatory T cells and absent in CD4−/CD8+ T cells. FOXP3 defects cause the scurfy phenotype in mice and IPEX syndrome in humans. Transcript variants encoding different isoforms have been identified. In human breast and colon cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
