The 6B11 monoclonal antibody reacts with an epitope of the CDR3 formed by the germ-line configuration of the V alpha 24 and J alpha 18 of the TCR alpha locus, which results in the expression of an invariant T-cell receptor (TCR) expressed by invariant NK T cells (iNKT).
This TCR is homologous to the murine V alpha 14J alpha 18 and pairs with restricted TCR beta chains, with V beta 11 being the most prominent in humans. NK T cells are restricted by CD1d and activated by CD1d-presented glycolipid ligands, leading to rapid IL-4, IL-13, and IFN-γ production.

Although iNKT cells exhibit NK-like cytolytic activity, they are key regulators of immune responses, producing both Th1 and Th2 cytokines in autoimmune disease, infection, and cancer. iNKT cell frequency in normal human peripheral blood varies from 0.01–1% of total T cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use at 5 µL (0.25 µg) per test in a final 100 µL volume. Cell count: 10⁵–10⁸ cells/test.

eFluor® 450 is an alternative to Pacific Blue®, excited by the Violet laser (405 nm) and emits at 445 nm. Ensure instrument compatibility.

Target Information

T cell receptors (TCRs) discriminate foreign from self-peptides presented by MHC class II molecules, critical for adaptive immunity. Recognition of self-peptides is linked to autoimmune disease, and mutant self-peptides are associated with tumors.
Superantigen engagement of TCRs is implicated in toxic shock syndrome. Autoantibodies to V beta segments occur in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially modulating inflammatory responses.

Most human T cells express TCR αβ with CD4 or CD8 (single positive). A minority are double negative (DN), which can increase in autoimmune and immunodeficiency disorders. γδ T cells, found mainly in epithelia, show limited TCR diversity and distinct antigen recognition, with subsets displaying antitumor and immunoregulatory functions.
