Target Information
EGF (Epidermal Growth Factor)는 170 kDa 단백질 키나아제인 EGF 수용체(EGFR)에 결합하여 작용을 발휘합니다. EGFR 활성화는 독성 환경 자극이나 EGF 결합 시 다양한 세포 경로를 유도하며, 수용체는 다른 가족 구성원과 호모 또는 헤테로다이머를 형성합니다. 각 이합체 수용체 복합체는 서로 다른 SH2 도메인 함유 단백질을 모집하여 고유한 신호 경로를 개시합니다.
EGF는 신장, 뇌, 전립선 및 침샘 등에 널리 발현되며, 강력한 유사분열 촉진 인자로 작용합니다. 인산화된 수용체는 GRB2와 같은 어댑터 단백질을 모집하여 RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, STAT 등 주요 신호 경로를 활성화합니다. 또한 NF-κB 신호 경로 활성화에도 관여할 수 있음이 연구로 제시되었습니다. EGF 유전자 결함은 저마그네슘혈증 4형의 원인이며, 발현 조절 이상은 특정 암의 성장 및 진행과 관련이 있습니다.
