Target Information

DISC1 (Disrupted in Schizophrenia 1) is a multifunctional protein that interacts with multiple proteins of the centrosome and cytoskeletal system. It plays a role in neuronal functions dependent on proper cytoskeletal regulation, including neuronal migration, neurite architecture, and intracellular transport.

DISC1 is expressed in various brain regions such as the hippocampus, lateral septum, amygdala, cerebral cortex, cerebellum, and paraventricular hypothalamus. Loss of DISC1 function leads to structural differences and is associated with schizophrenia through chromosomal translocations.

The DISC1 protein contains a globular N-terminal domain and a helical C-terminal domain, both capable of interacting with other proteins. It associates with proteins involved in the centrosome and cytoskeletal system (MIPT3, MAP1A, NUDEL), membrane receptor localization (alpha-actinin 2, beta 4-spectrin), and signal transduction (ATF-4, ATF-5).

DISC1 is implicated in intracellular transport, neurite architecture, and neuronal migration, processes thought to be pathogenic in schizophrenia. Reduced DISC1 expression in neurons accelerates neuronal integration and causes abnormal morphological development, suggesting a role in dendritic development and synapse formation.
