Target Information
The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR acts as a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
The AR signaling pathway is crucial for development and function of male reproductive organs such as the prostate and epididymis, and also plays roles in nonreproductive tissues including muscle, hair follicles, and brain. AR is a phosphoprotein involved in MAP kinase regulation; inhibition of the MEK1/2 pathway affects AR phosphorylation. Dysregulation of AR signaling is associated with diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene cause complete androgen insensitivity (CAIS).
