Product Specific Information

Description:
The monoclonal antibody HP-F1 recognizes CD85j (also known as ILT2, LILRB1, LIR1). CD85j is a member of the ILT/LIR/MIR family, a single transmembrane glycoprotein with a cytoplasmic domain containing four ITIMs that signal via SHP-1.
Expression occurs on myeloid cells (monocytes, dendritic cells) and subsets of NK, T, and B cells. It is associated with leukemias such as ALL and CLL and plays roles in viral infections (HIV, EBV, CMV).
Ligands include MHC Class I molecules (HLA-G, A, F, B27, E, F). HP-F1 reduces CD16-dependent cytolytic activity of NK cells.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for human peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

Blocking Buffer Usage:
Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.

• Add 5 µL per sample before combining antibody cocktails with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at 19q13.4.
It encodes a receptor with extracellular immunoglobulin domains, a transmembrane domain, and cytoplasmic ITIM motifs.
Expressed on immune cells, it binds MHC class I molecules and transduces inhibitory signals to regulate immune responses, inflammation, and cytotoxicity.
Multiple transcript variants encoding different isoforms are known.
