Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j, also known as ILT2, LILRB1, and LIR1. CD85j is a member of the ILT/LIR/MIR family and is a single transmembrane glycoprotein with a long cytoplasmic domain containing four ITIMs that signal through SHP-1 interactions.

Expression is found on myeloid cells (monocytes and dendritic cells) and subsets of NK, T, and B lymphoid cells. It is correlated with leukemias such as ALL and CLL, and plays an important role in viral infections including HIV, Epstein-Barr, and CMV.

Ligands for CD85j include MHC Class I molecules such as HLA-G, A, F, B27, E, and F. HP-F1 antibody has been shown to reduce CD16-dependent cytolytic activity of NK cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test (in 100 µL final volume).

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation. Add 5 µL per sample for optimal staining. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, featuring engineered fluorescent signatures designed to minimize spillover and spread impacts.

Target Information

The CD85j gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosome region 19q13.4. It encodes a receptor with two to four extracellular immunoglobulin domains, a transmembrane domain, and multiple ITIMs. This receptor is expressed on immune cells, binds MHC class I molecules, and transduces inhibitory signals to regulate immune responses, inflammation, and cytotoxicity. Multiple transcript variants exist for this gene.
