Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (ILT2), also known as LILRB1 and LIR1.
CD85j is a member of the ILT/LIR/MIR receptor family and is a single transmembrane glycoprotein containing four ITIMs that signal through SHP-1.
It is expressed on myeloid cells (monocytes, dendritic cells) and subsets of lymphoid cells (NK, T, B cells).

Expression is correlated with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV).
Ligands include MHC Class I molecules (HLA-G, A, F, B27, E, F).
HP-F1 antibody reduces CD16-dependent NK cell cytolytic activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells at 5 µL (0.25 µg)/test.
A test is defined as the antibody amount to stain a 100 µL cell sample (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining antibody cocktails with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

The CD85j gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosomal region 19q13.4.
This receptor contains extracellular immunoglobulin domains, a transmembrane domain, and ITIM motifs.
It binds MHC class I molecules on antigen-presenting cells, transmitting inhibitory signals to regulate immune activation and prevent autoreactivity.
Multiple transcript variants encoding distinct isoforms are known.
