The monoclonal antibody eBio2B2-G20 reacts with human IL-21, a potent stimulator of NK and cytolytic T cells. IL-21 is a 131 amino acid protein closely related to IL-2 and IL-15, expressed in activated CD4+ cells.
IL-21 stimulates proliferation of B cells via CD40 crosslinking and bone marrow progenitor cells, and enhances NK-cell marker CD56 expression in the presence of IL-15.

• Reported for use in ELISA detection of IL-21.

Applications Tested:

• Tested as detection antibody in sandwich ELISA for human IL-21 analysis with purified eBio3A3-N2 (Product #14-7219-82) as capture antibody and recombinant human IL-21 (Product #14-8219) as standard.
• Recommended ELISA concentration range: 0.5–2 µg/mL.
• Include standard curve with recombinant IL-21 range 4000 pg/mL–60 pg/mL per plate.
• Post-manufacturing filtration: 0.2 µm.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine mainly produced by NKT, Th17, and TFH cells.
In TFH cells, IL-21 triggers autocrine signaling via IL-21R and STAT3, activating transcription factor Bcl6—critical for TFH development and effector function.
IL-21 promotes T cell-dependent B cell differentiation, IgG production, and induces apoptosis in naive B cells.
In Th17 cells, IL-21 signaling via STAT3, IRF4, and RORγt upregulates IL-23R, supporting maturation and maintenance by IL-23.
IL-21 also downregulates Foxp3, influencing immune balance.
IL-21 deficient mice are resistant to chemically induced colitis due to impaired Th17 response, whereas elevated IL-21 levels are observed in colitis models.
