The affinity-purified polyclonal rabbit antibody reacts with mouse and rat tumor necrosis factor-alpha (TNFα). Mouse TNFα is a 17 kDa factor produced by macrophages, monocytes, neutrophils, CD4 T cells, and NK cells, with a 26 kDa membrane-bound form. TNFα is cytolytic and plays a key role in immune regulation. Bioactive TNFα exists as a homotrimer.

Applications Reported: ELISA and ELISPOT.

Applications Tested:
This biotinylated rabbit anti-mouse/rat TNFα polyclonal antibody is tested as the detection antibody in a sandwich ELISA for mouse or rat TNFα, used with the monoclonal capture antibody clone TN3.19 (14-7423) and recombinant TNFα (14-8321) as standard.
Recommended detection range: 0.5–2 µg/mL.
Standard curve range:

• Mouse TNFα: 2000–15 pg/mL
• Rat TNFα: 5000–30 pg/mL

Recognition of mouse TNFα is not interfered with by 1000x excess soluble TNFα receptor type I or II.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNFα regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.

It exists as multimers (dimers, trimers, or pentamers) and shows a 17 kDa band under non-reducing SDS-PAGE conditions. Mouse knockout studies indicate neuroprotective roles, and TNFα induces tumor necrosis in tumor-bearing mice. It is involved in immune responses to bacterial, viral, parasitic, and fungal infections and plays a role in tumor necrosis. TNFα acts synergistically with interferon-gamma and has been implicated in autoimmune diseases, insulin resistance, and cancer.
