Target Information
The PKC family of serine/threonine kinases, including PRKCG (PKC gamma), is activated intracellularly by signal transduction pathways. In humans, at least 12 different PKC polypeptides have been identified, differing in structure, tissue distribution, localization, and substrate specificity. PKC alpha, beta I, beta II, and gamma form the conventional family, and their activities are Ca²⁺- and phospholipid-dependent.
Protein kinase C can be activated by calcium and diacylglycerol, and PKC family members phosphorylate various protein targets involved in diverse signaling pathways. PKC also serves as a major receptor for phorbol esters, a class of tumor promoters. PKC gamma is expressed exclusively in the brain and spinal cord, localized to neurons, and plays roles in neuronal functions such as long-term potentiation (LTP) and long-term depression (LTD). Knockout studies suggest involvement in neuropathic pain development. Defects in PKC gamma are associated with spinocerebellar ataxia-14 (SCA14).
