The AFL4 monoclonal antibody reacts with mouse VEGF receptor-3 (Flt-4), a 195 kDa endothelial-specific receptor tyrosine kinase. During embryogenesis, all endothelial cells express VEGFR-3, but in adults, expression is limited to lymphatic endothelium. VEGFR-3 expression can be re-induced in adult tissues during tumor growth, indicating a role in tumor angiogenesis. VEGF-C and VEGF-D bind and activate VEGFR-3. AFL4 functions as an antagonist monoclonal antibody.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested in flow cytometry using in vitro differentiated mouse endothelial cells. Recommended ≤0.5 µg/test (100 µL final volume). Optimal titration required for specific assays.
Target Information
Vascular endothelial growth factor (VEGF) regulates embryonic blood vessel development and adult angiogenesis. VEGF-C specifically stimulates lymphatic vessel growth via VEGFR-3. Inactivation of VEGFR-3 in mice leads to abnormal vessel organization and embryonic cardiovascular failure, demonstrating its essential role in vascular and lymphatic development.
