Product Specific Information

The SN3 monoclonal antibody reacts with the human CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa molecule is GPI-anchored and expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. P-selectin (CD62P) has been reported to bind CD24.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for use at 5 µL (0.4 µg) per test on normal human peripheral blood cells. A test corresponds to staining in 100 µL final volume with 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., Propidium Iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment with NovaFluor dyes.
• The conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining performance.
• Add 5 µL CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls, use 5 µL buffer per 100 µL cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescence signatures and reduced spectral spillover.

Target Information

CD24 is a GPI-anchored glycoprotein expressed during various stages of B-cell development, from pro-B cells to mature surface Ig-positive B cells. Expression is low or absent in plasma cells. It is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and non-Hodgkin’s lymphomas. CD24 is implicated in B-cell proliferation, maturation, and autoimmunity regulation.
