Product Specific Information

The SN3 monoclonal antibody reacts with the human CD24 molecule (Heat Stable Antigen, HSA). This 35–50 kDa molecule is GPI-anchored and expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. It has been reported that P-selectin (CD62P) binds to CD24.

Applications Tested:

• Pre-titrated and tested by flow cytometry on normal human peripheral blood cells
• Recommended use: 5 µL (0.6 µg) per test (final volume 100 µL; 10⁵–10⁸ cells/test)

• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD24 is a GPI-anchored glycoprotein expressed during multiple stages of B-cell development, from bone marrow pro-B cells to mature B cells. Expression is low or absent in plasma cells. CD24 is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
