Product Specific Information

The 22URTI monoclonal antibody reacts with human interleukin (IL)-22, a 20 kDa cytokine of the IL-10 family secreted primarily by Th17 and NK cells. IL-22 expression is induced more strongly by IL-23 than by IL-6 or TGF-β, suggesting secretion by fully differentiated Th17 cells. The IL-22 receptor is a heterodimer of IL-10R2 and IL-22R1.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested for intracellular staining of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).

Use 5 µL (0.25 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.

Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is suitable for cytometers equipped with a 355 nm UV laser.
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.

Target Information

IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine and a member of the IFN-IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29). It is produced mainly by activated T cells and NK cells. The IL-22 gene is located on chromosome 12q and structurally related to IL-10.

IL-22 acts via a receptor complex of IL-22R1 and IL-10R2, activating the JAK-STAT pathway. A soluble receptor, IL-22BP, antagonizes IL-22 binding. IL-22 targets nonimmune cells (skin, digestive, respiratory tissues, hepatocytes, keratinocytes) and regulates genes associated with innate immunity of the skin. It promotes host defense, wound healing, and inflammation via STAT3 activation.
