Product Specific Information

The eBio64DEC17 antibody reacts with human IL-17A and is a neutralizing antibody. IL-17A is a CD4+ T cell-derived cytokine involved in inflammatory responses and elevated in diseases such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.

Human IL-17A exists as a glycosylated 20–30 kDa homodimer produced by activated CD4+ T cells. It enhances ICAM-1 expression in fibroblasts and induces IL-6, IL-8, G-CSF, and PGE2 secretion from epithelial, endothelial, or fibroblastic cells. IL-17A also supports CD34+ progenitor proliferation and neutrophil maturation.

Th17 cells, which produce IL-17A, develop via an IL-23-dependent pathway distinct from Th1 and Th2 differentiation. IFN-γ and IL-4 negatively regulate Th17 generation.

This antibody identifies the same intracellular IL-17A population as eBio64CAP17 and is validated for intracellular staining followed by flow cytometric analysis. It is pre-diluted and optimized for use with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

Recommended Use:

  • Use 5 µL (1.0 µg) per test (100 µL total volume)
  • Cell count: 10⁵–10⁸ cells/test
  • Compatible with ultraviolet (355 nm) laser cytometers

Notes:

  • Use Brilliant Stain Buffer™ or Super Bright Complete Staining Buffer to minimize non-specific interactions when combining multiple polymer dyes.
  • Protect from light due to photo-induced oxidation sensitivity.
  • Fixed samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer or 1-step Fix/Lyse Solution.

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a cytokine produced by CD4+ T cells that promotes inflammation and is elevated in several autoimmune and inflammatory diseases. IL-17A is a 32 kDa disulfide-linked homodimer that signals through IL-17R/CDw217. It induces ICAM-1 expression and stimulates IL-6, IL-8, G-CSF, and PGE2 secretion. IL-17A activates NF-κB and MAPK pathways and enhances COX-2 and nitric oxide production.
