Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios.
Helios is a zinc-finger transcription factor related to Ikaros, expressed in hematopoietic stem cells and highly in regulatory T cells, but not in mature B cells, dendritic cells, or myeloid cells. It forms homo- or heterodimers with other Ikaros family members to regulate transcription.
Eight isoforms exist, some with dominant-negative functions, upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 has been reported to cross-react with rat Helios.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated for intracellular staining of mouse lymph node cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Recommended use: 5 µL (0.125 µg) per test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is designed for use with UV (355 nm) lasers. Verify instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce non-specific interactions.

Light Sensitivity:
Protect from light; dye is sensitive to photo-induced oxidation.

Fixation:
Samples can be stored with IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

Helios is a member of the Ikaros family of zinc-finger proteins (Ikaros, Aiolos, Helios), which are hematopoietic-specific transcription factors regulating lymphocyte development. The protein forms homo- or heterodimers with other Ikaros family members and functions in early hematopoietic development. Multiple transcript variants exist, though some have uncertain biological validity.
