Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE is present at low levels in serum and plays a key role in allergic responses by binding to mast cells and basophils via the Fc epsilon receptor I, triggering histamine release. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.2 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer Usage:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails before mixing with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

IgE is one of five immunoglobulin classes in mammals. It consists of two epsilon heavy chains and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on basophils and mast cells. Crosslinking induces degranulation and release of allergic mediators. IgE also contributes to defense against parasitic infections.
