Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. Natural IgE levels in serum are low and play a key role in allergy responses by binding to mast cells and basophils via Fc epsilon receptor I, triggering histamine release. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms, both recognized by IgE21.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.2 µg) per test in a final volume of 100 µL. Cell numbers may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, add 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

IgE is one of five mammalian immunoglobulin classes. It consists of two epsilon heavy chains and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation, promoting allergic responses. IgE also contributes to immunity against parasites.
