Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms of Ki-67 (345 and 395 kDa) are expressed in dividing cells and absent in resting cells (G0). During interphase, Ki-67 localizes to the nucleus, redistributing to chromosomes during mitosis, and associates with heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 expression serves as a marker for the fraction of proliferating cells.

This antibody also recognizes canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis, immunohistochemistry, and immunocytochemistry.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometric analysis using Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Use at 5 µL (0.06 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue laser.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus through its FHA domain.
Ki-67 expression varies by cell cycle stage: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase.
It serves as a proliferation marker, undetectable in G0 cells and increasing from G1 through mitosis.
Ki-67 antibodies are useful for assessing the growth fraction in neoplasms, with prognostic value comparable to thymidine-labelling index.
Commonly used as a neuronal marker of cell cycling and proliferation.
