Product Specific Information
The Y-Ae antibody reacts with a class II MHC self Ea peptide (peptide 52–68) bound to I-Ab molecules. It detects a determinant expressed on a subset of class II I-Ab molecules in strains that also express class II I-Eb. This determinant is found on peripheral B cells and thymic medulla cells, but not on thymic cortical epithelium. It is also expressed on other antigen-presenting cells such as macrophages and dendritic cells, but not on invariant chain-associated class II MHC complexes.
The reactivity of the Y-Ae antibody is MHC-restricted. It was generated by immunizing [B10.MBR x B10.D2]F1 recipients with B10A(5R) LPS blasts. The antibody reacts with 10–15% of surface I-Ab molecules in strains expressing I-Eb (e.g., B10.A(5R), B10.A(3R)). It does not react with I-A molecules from strains lacking functional I-E genes (e.g., B6, B10) or with mutant I-A molecules. The Y-Ae antibody recognizes Ea peptide (Ea 52–68) bound to I-Ab, but not when bound to I-Ak or I-Abm-12.

Applications Tested: Tested on mouse peripheral blood cells at ≤0.5 µg/test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information
MHC class II molecules are heterodimers composed of non-covalently associated alpha (31–34 kDa) and beta (26–29 kDa) chains. Antigen presentation requires lysosomal proteases to process endocytosed antigens into peptides and to remove the invariant chain (Ii) blocking the peptide-binding site. MHC class II molecules present exogenous antigens to CD4⁺ T lymphocytes (T helper cells). CD4 interacts with non-polymorphic residues of MHC class II.
HLA and MHC antibodies are essential tools in Immunopeptidomics, aiding in the identification and characterization of neoantigens via high-performance liquid chromatography and tandem mass spectrometry.
