This MRRBY monoclonal antibody recognizes human and mouse mammalian target of rapamycin (mTOR, FRAP, RAFT) phosphorylated on S2448.
mTOR is a serine/threonine kinase that regulates cell growth, proliferation, motility, survival, protein synthesis, and transcription.
It acts as an ATP and amino acid sensor, balancing nutrient availability with cellular metabolism.
Phosphorylation at S2448 occurs via the PI3K/Akt signaling pathway and is linked to tumorigenesis, making mTOR a key target in cancer research.
Applications Reported:
- Intracellular staining followed by flow cytometric analysis.
Applications Tested:
- Pre-titrated for intracellular staining of human peripheral blood cells.
- Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).
Staining Protocols:
- Protocol A: Two-step for cytoplasmic proteins (flexible for surface and intracellular detection).
- Protocol B: One-step for nuclear proteins (ideal for transcription factors).
- Protocol C: Two-step Fixation/Methanol for phospho-specific signaling discrimination.
