Product Specific Information
The ALYSCLN monoclonal antibody reacts with human IRF5, a 60–63 kDa member of the interferon regulatory factor (IRF) family of transcription factors involved in host defense against viruses. IRF5 regulates IFN-α and IFN-β and induces pro-inflammatory cytokines such as IL-6, IL-12, and TNF-α in response to TLR signaling. MyD88 and TRAF6 interact directly with IRF5, promoting its nuclear translocation upon TLR activation.
IRF5 also plays roles in cell cycle regulation and apoptosis, both p53-dependent and independent. Genetic polymorphisms in the IRF5 locus are associated with autoimmune diseases such as Sjögren’s syndrome, inflammatory bowel disease, multiple sclerosis, and systemic lupus erythematosus.
Recent studies show IRF5 involvement in M1 macrophage polarization, upregulating IL-12 and IL-23 while repressing IL-10. IRF5 is highly expressed in monocytes, macrophages, and plasmacytoid dendritic cells, and also found in B and T cells.
Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometry using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00). Recommended use at 5 µL (0.25 µg) per test (100 µL final volume). Cell count per test: 10⁵–10⁸ cells.

eFluor® 660 replaces Alexa Fluor® 647. It emits at 659 nm and is excited with a 633 nm red laser.

Target Information
IRF5 is part of the interferon regulatory factor family, transcription factors involved in interferon activation and immune modulation. It contains a conserved N-terminal DNA-binding domain with tryptophan repeats. Alternative splice variants encode different isoforms.
