Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and interacts with protein tyrosine kinase p56lck, playing a key role in T-cell development and activation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of human peripheral blood leukocytes. Recommended usage ≤0.125 µg/test (100 µL volume). Cell number can range from 10⁵–10⁸ cells/test. Optimize titration for best results.
Alexa Fluor® 700 emits at 723 nm and is excited by the red laser (633 nm). Recommended optical setup: 685 LP mirror and 710/20 filter.

Target Information
Cluster of Differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential for signal transduction and T-cell differentiation/activation. CD8 acts as a co-receptor with TCR for MHC-I molecules. It exists as CD8αβ heterodimers or CD8αα homodimers on T cells, NK cells, monocytes, and dendritic cells. Binding of MHC-I/peptide complexes triggers Lck recruitment, leading to lymphokine production and cytotoxic T lymphocyte activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
