Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by a majority of thymocytes, a subpopulation of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays an important role in T-cell development and optimal functioning of mature T cells. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse thymocyte and splenocyte suspensions. Recommended ≤0.125 µg/test. A test is defined as the amount of antibody that will stain cells in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Alexa Fluor® 700 emits at 723 nm and can be excited with a 633 nm red laser. Most instruments require a 685 LP mirror and 710/20 filter.

Target Information

The CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. CD4 is primarily expressed in T helper cells, but also found in monocytes, macrophages, and dendritic cells. At the tissue level, CD4 expression is detected in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 functions in early T-cell activation through association with the T-cell receptor complex and tyrosine kinase Lck. It may also mediate neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
