Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Formalin-fixed tissues require heating in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
The DAXX gene encodes a multifunctional protein localized in both the nucleus and cytoplasm. It interacts with various proteins such as apoptosis antigen Fas, centromere protein C, and transcription factor E26 (ETS family).
In the nucleus, DAXX acts as a transcriptional repressor binding to sumoylated transcription factors, with repression relieved upon sequestration into promyelocytic leukemia nuclear bodies or nucleoli.
DAXX also associates with centromeres during G2 phase and may regulate apoptosis in the cytoplasm. Its localization and function are modulated by post-translational modifications such as sumoylation, phosphorylation, and polyubiquitination.
Alternative splicing produces multiple transcript variants.
