The MHR73-11 monoclonal antibody reacts with human CD180 (RP105), a 105 kDa type I transmembrane molecule of the Toll-like receptor (TLR) family. It features an extracellular leucine-rich repeat domain and a cytoplasmic region homologous to the type I IL-1 receptor.
RP105 associates with MD-1 and is expressed on B cells, monocytes/macrophages, and dendritic cells. It is mainly found on mature B cells in mantle zones. The RP105/MD-1 complex, together with TLR4, mediates B cell recognition and signaling of LPS.

MHR73-11 activates B cells, increasing cell size, CD80 expression, and DNA synthesis, and protects against irradiation- or dexamethasone-induced apoptosis. RP105 plays a role in regulating B cell growth and death. In SLE patients, RP105-negative B cells are associated with disease activity.

Applications Reported: Flow cytometry and in vitro functional studies.
Applications Tested: ≤1 µg/test for flow cytometry (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.
Storage and Handling: Use in sterile environment.

Target Information

CD180 (RP105, Bgp95, LY64) is a type I membrane glycoprotein of the Toll-like receptor family. It lacks the TIR domain and depends on MD-1 coexpression. CD180 regulates LPS recognition and signaling in B cells by interacting with the TLR4 signaling complex, inhibiting microbial ligand binding.

Ligation of CD180 induces B cell activation, CD80/CD86 upregulation, and increased cell size. It consists of extracellular leucine-rich repeats (LRR) and a short cytoplasmic tail, forming the RP105/MD-1 complex.
CD180 dysfunction is associated with Kawasaki disease and may contribute to rheumatic disease pathology.
