Product Specific Information

The 6C2 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2). Mouse TLR2 is expressed by the myeloid lineage, including macrophages and dendritic cells in splenocytes and the RAW264.7 cell line.
TLR2 and TLR4 act as pattern recognition receptors and signaling molecules in response to bacterial lipoproteins, playing key roles in innate immunity and inflammation.

Applications Reported:
6C2 has been reported for use in flow cytometric analysis. For optimal staining, directly conjugated 6C2 (FITC, PE, or Biotin) is recommended in combination with FcR blocking using anti-mouse CD16/CD32 (clone 93, Product #14-0161).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg per test (defined as staining a cell sample in 100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

TLR2 is a member of the Toll-like receptor family, essential for pathogen recognition and activation of innate immunity.
It acts through adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKCα/β and NF-κB.
TLR2 forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins.
It is highly expressed in peripheral blood leukocytes and mediates responses to Gram-positive bacteria and yeast.
Expression is regulated by immune modifiers: LPS, GM-CSF, IL-1, and IL-10 upregulate TLR2, while IL-4, IFN-γ, and TNF downregulate expression in monocytes.
