Description:
The mT2.7 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2). Mouse TLR2 is expressed by the myeloid lineage, including macrophages and dendritic cells in splenocytes and the RAW264.7 cell line. The Toll family, consisting of at least twelve members in humans and mice, are type I transmembrane proteins characterized by an extracellular leucine-rich repeat domain and a cytoplasmic domain homologous to the type I IL-1 receptor. TLR2 and TLR4 act as pattern recognition receptors and signaling molecules in response to bacterial lipoproteins, playing key roles in innate immunity and inflammation.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.5 µg per test (100 µL final volume). Optimal performance should be determined by titration.

Target Information
TLR2 (Toll-like receptor 2) is part of the TLR family, crucial for pathogen recognition and activation of innate immunity. TLR2 interacts with adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKCα/β and NF-κB. It forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins.
TLR2 is highly expressed in peripheral blood leukocytes and mediates responses to Gram-positive bacteria and yeast. Expression varies by cell type and can be modulated by immune factors such as LPS, GM-CSF, IL-1, IL-10 (upregulation), and IL-4, IFN-γ, TNF (downregulation).
