Product Specific Information

This SRBCZX monoclonal antibody recognizes STAT5 phosphorylated on tyrosine 694. STAT proteins are activated by ligand binding to cytokine receptors associated with JAK kinases. Phosphorylated STAT5 translocates to the nucleus to regulate gene transcription.

Phosphorylation of STAT5 on Tyr694 is essential for dimer formation, nuclear translocation, and DNA binding. Specificity of SRBCZX clone was confirmed by ELISA and flow cytometry.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested on stimulated U937 cells. Use at 5 µL (1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Recommended Protocol:
Use Protocol C (Two-step Fixation/Methanol). Protocol A and B are not suitable for this target.

Target Information

STAT5 alpha is a transcription factor activated by cytokines and growth factors through receptor-associated kinases. It forms dimers that translocate to the nucleus to activate transcription. STAT5 alpha mediates responses to IL2, IL3, IL7, GM-CSF, erythropoietin, and others. It plays critical roles in reproduction, lactation, immune function, and growth.

Activation of STAT5 alpha via TEL/JAK2 fusion is linked to tumorigenesis. The mouse homolog induces BCL2L1/BCL-X(L) expression, suggesting anti-apoptotic activity. STAT5 alpha and beta share 96% sequence similarity and are co-expressed in most tissues.
