Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins. It inhibits amyloid fibril formation by various proteins (APP, APOC2, B2M, CALCA, CSN3, SNCA, LYZ variants) in vitro, acting in an ATP-independent manner. Clusterin maintains partially unfolded proteins for refolding by other chaperones such as HSPA8/HSC70 but does not refold proteins itself.

Binding to cell surface receptors triggers internalization of the chaperone-client complex and subsequent lysosomal or proteasomal degradation. The secreted isoform protects cells against apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ubiquitin-protein ligase complexes, promoting ubiquitination and degradation of target proteins such as COMMD1 and IKBKB, thereby modulating NF-kappa-B activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis. Clusterin plays a critical role in regulating cell proliferation.
