Product Specific Information

The R1-2 monoclonal antibody reacts with mouse CD49d, the 150 kDa integrin alpha4 subunit.
CD49d forms a non-covalent complex with integrin beta1 (CD29), known as VLA-4, which acts as a receptor for fibronectin and VCAM-1 (CD106).
This complex is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils.
CD49d also associates with integrin β7 and binds to the mucosal vascular addressin (MAdCAM-1).

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested with mouse splenocytes; optimal performance at ≤0.5 µg/test (100 µL final volume).
Cell number may vary (10⁵–10⁸ cells/test); titration recommended for best results.

Target Information

Integrin alpha 4 (CD49d, ITGA4) is a member of the integrin alpha chain family.
Integrins are heterodimeric membrane proteins composed of alpha and beta chains.
Alpha4 associates with beta1 or beta7 chains, and ligand-binding sites of both integrin α4β1 and α4β7 are located on the alpha4 chain.
Ligands include MAdCAM, VCAM, and fibronectin.
MAdCAM is the principal ligand for integrin α4β7.
Recent studies show that HIV-1 envelope can mimic MAdCAM by binding and signaling through integrin α4β7, the gut mucosal homing receptor for peripheral T cells.
