Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4⁺ and CD8⁺ peripheral T lymphocytes and B cells.
CD45RA⁺ T cell populations are mainly naive/virgin, allowing the use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Applications Tested: Tested on normal human peripheral blood cells at ≤ 0.5 µg/test (100 µL final volume).
Cell number should be empirically determined (10⁵–10⁸ cells/test).
Careful titration is recommended for optimal performance.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid subtypes.
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of all lymphocyte surface proteins.
It regulates Src-family kinases, crucial in lymphocyte development and antigen signaling.
CD45 isoforms differ due to alternative splicing, varying in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
Expression depends on cell type and physiological state.
CD45 also promotes cell survival via integrin-mediated signaling and is involved in DNA fragmentation during apoptosis.
It is expressed on all hematopoietic cells except mature red cells.
CD45 dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
