Casein kinase II (CK2) is a protein serine/threonine kinase found in all eukaryotic cells, indicating fundamental cellular functions.
The holoenzyme is a tetramer containing two alpha or alpha-prime subunits (or one of each) and two beta subunits. The alpha subunit acts as the catalytic unit, while the function of beta subunits remains unclear.
CK2 can phosphorylate several intracellular signaling proteins involved in tumor suppression (p53, PTEN) and tumorigenesis (myc, jun, NF-κB). It also influences Wnt signaling via β-catenin phosphorylation and PI3K signaling via Akt phosphorylation.
