This antibody detects endogenous levels of Caspase 8 only when phosphorylated at Serine 347.
Target Information
Caspase 8 (CASP8) is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution phase of apoptosis. Caspases exist as inactive proenzymes composed of a pro-domain, a large protease subunit, and a small protease subunit. Activation requires proteolytic processing at conserved internal aspartic residues to form a heterodimeric enzyme.
Caspase 8 is involved in programmed cell death induced by Fas and various apoptotic stimuli. Its N-terminal FADD-like death effector domain suggests interaction with Fas-interacting protein FADD. Caspase 8 has been detected in the insoluble fraction of affected brain regions from Huntington disease patients, suggesting a role in neurodegenerative diseases. Activated Caspase 8 triggers downstream caspases such as Caspase 9, committing the cell to apoptosis. It also reacts with Jurkat cells and tonsil tissues. Overexpression induces apoptosis, which can be inhibited by ICE family-specific inhibitors. Multiple alternatively spliced transcript variants exist, though not all have been fully characterized.
