Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR receptor family. CD85j is a single transmembrane glycoprotein with four ITIMs that signal through SHP-1. It is expressed on myeloid cells (monocytes, dendritic cells) and subsets of NK, T, and B cells. Expression is associated with leukemias (ALL, CLL) and viral infections such as HIV, EBV, and CMV.

• HP-F1 antibody reduces CD16-dependent NK cell cytolytic activity

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining
  • Add 5 µL per sample (regardless of number of NovaFluors)
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

The gene encoding CD85j belongs to the leukocyte immunoglobulin-like receptor (LIR) family located on chromosome 19q13.4. This receptor contains extracellular Ig domains, a transmembrane region, and cytoplasmic ITIM motifs. It binds MHC class I molecules on antigen-presenting cells and transduces inhibitory signals, regulating immune response and preventing autoreactivity. Multiple transcript variants exist for this gene.
