Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR receptor family.
CD85j is a single transmembrane glycoprotein with a long cytoplasmic domain containing four ITIM motifs that signal via SHP-1.
Expression is found on myeloid cells (monocytes and dendritic cells) and subsets of NK, T, and B lymphocytes.
It is correlated with leukemias (ALL, CLL) and viral infections such as HIV, EBV, and CMV.
Ligands include MHC Class I molecules (HLA-G, A, F, B27, E, F).
HP-F1 antibody reduces CD16-dependent NK cell cytolytic activity.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry on normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimal spillover.

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosomal region 19q13.4.
It encodes a receptor with extracellular Ig domains, a transmembrane domain, and ITIM motifs mediating inhibitory signaling.
Expressed on immune cells, it binds MHC class I molecules on antigen-presenting cells, suppressing immune activation.
It regulates inflammation and cytotoxicity to limit autoreactivity.
Multiple transcript variants encoding different isoforms are known.
