Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j, also known as ILT2, LILRB1, and LIR1. CD85j is a member of the ILT/LIR/MIR family and is a single transmembrane glycoprotein with four ITIMs that signal through SHP-1. Expression occurs on myeloid cells (monocytes, dendritic cells) and subsets of lymphoid cells (NK, T, B cells). Correlated with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV). Ligands include MHC Class I molecules such as HLA-G, A, F, B27, E, and F. HP-F1 antibody reduces CD16-dependent NK cell cytolytic activity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor Compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample or per 100 µL of cell suspension (10³–10⁸ cells). Recommended for antibody cocktails before mixing with cells. Cat. No. C001T06F01.

NovaFluor Technology:
Based on Phiton™ technology using nucleic acid dye structures engineered for optimized fluorescence with reduced spillover and spread.
Excitation: 494 nm; Emission: 585 nm; Laser: 488 nm (Blue).

Target Information

CD85j gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at 19q13.4. It encodes a receptor with extracellular Ig domains, a transmembrane region, and ITIMs. Expressed on immune cells, it binds MHC class I molecules and transduces inhibitory signals to regulate immune response, inflammation, and cytotoxicity. Multiple transcript variants exist.
