Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 세포사멸, 분화 등 다양한 세포 과정에 관여합니다. SMAD2는 TGF-beta 수용체와 상호작용하며 핵 내로 이동하여 TGF-beta 신호전달의 중심적 역할을 수행합니다. ERK1에 의해 MH1 도메인의 칼모듈린 결합 영역 내 Thr8이 인산화되면 SMAD2의 전사 활성화가 증가하며, 이는 칼모듈린에 의해 음성 조절됩니다. TGF-beta 신호에 반응하여 SMAD2는 수용체에 의해 인산화되고, SARA와의 결합이 해제되며 SMAD4와 결합하여 핵으로 이동해 전사 억제 복합체를 형성합니다. 또한 activin type 1 receptor kinase에 의해 인산화되어 activin 신호를 매개할 수 있습니다. SMAD2 단백질을 암호화하는 대체 스플라이싱 전사체 변이도 보고되었습니다.
