Target Information

BID is a death agonist that heterodimerizes with either agonist BAX or antagonist BCL2. It belongs to the BCL-2 family of cell death regulators. BID mediates mitochondrial damage induced by caspase-8 (CASP8); CASP8 cleaves BID, and the COOH-terminal fragment translocates to mitochondria to trigger cytochrome c release.
In living cells, BID exists in an inactive form in the cytosol and becomes activated by caspase-8 in response to TNF-alpha or Fas ligand. The cleaved C-terminal 15 kDa fragment (p15) localizes to mitochondria, initiating cytochrome c release and apoptosis.
BID acts as a direct molecular link between caspase-8 activation and mitochondrial death machinery.
