Target Information

The SMAD1 protein belongs to the SMAD family, homologous to the Drosophila mothers against decapentaplegic (Mad) and C. elegans Sma genes. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways.

SMAD1 mediates bone morphogenetic protein (BMP) signaling, which regulates cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, SMAD1 is phosphorylated and forms a complex with SMAD4 to regulate transcription. It is also targeted by SMAD-specific E3 ubiquitin ligases (SMURF1, SMURF2) for ubiquitination and proteasomal degradation. Alternatively spliced variants encoding the same protein have been observed.
