Product Specific Information

The LN3 monoclonal antibody reacts with human MHC class II molecule HLA-DR, expressed on antigen presenting cells such as B cells, monocytes, macrophages, dendritic cells, and activated T cells.
HLA-DR is a heterodimeric transmembrane protein composed of alpha and beta subunits, essential for peptide presentation to CD4⁺ T lymphocytes.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-titrated and validated on normal human peripheral blood cells.
    Recommended use: 5 µL (0.125 µg) per test (100 µL final volume).
    Cell number range: 10⁵–10⁸ cells/test.
  • Fluorochrome Information: eFluor® 450 emits at 445 nm, excited at 405 nm Violet laser.
    Ensure instrument compatibility with this fluorochrome.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The alpha chain gene (DRA) has 5 exons, encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail.
DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, while DRB1 alleles define peptide binding specificity.
HLA-DR is expressed mainly on antigen presenting cells (B lymphocytes, monocytes, macrophages, thymic epithelial cells, activated T lymphocytes).
MHC class II molecules (DR, DQ, DP) present processed peptides to T-helper cells, initiating immune responses.
HLA and MHC antibodies are crucial in Immunopeptidomics, aiding neoantigen identification and characterization via LC-MS/MS.
