Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a member of the Gata family of transcription factors. Gata-3 is a T cell-specific transcription factor important for thymic development and Th2 differentiation. Expression during embryonic development occurs in the central nervous system, skin, mammary glands, and kidney. Homozygous knockout of Gata-3 is embryonic lethal, highlighting its essential developmental role.

Gata-3 is also critical for T cell commitment and survival, primarily expressed in CD4 single-positive thymic cells. During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.

Alternative splice variants have been reported, particularly in the MCF7 cell line. The TWAJ antibody recognizes both 50 and 45 kDa forms of the protein.

Staining requires the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Crossreactivity in rhesus monkeys has been published.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Prediluted and tested by intracellular staining of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
eFluor 450 is an alternative to Pacific Blue. It emits at 446 nm and is excited with the violet laser (405 nm). Ensure instrument compatibility.

Target Information

The genes for all four subunits of the T-cell antigen receptor (α, β, γ, δ) are controlled by distinct enhancers and their binding proteins. GATA3 binds to enhancer elements of all four TCR genes, acting as a zinc finger transcription factor. GATA3 mRNA is expressed in T cells but not in B cells or macrophages, indicating its role in T-cell lineage regulation and TCR gene activation.

The human GATA3 gene comprises six exons over 17 kb of DNA. Its two zinc fingers are encoded by separate exons highly conserved with GATA1, though otherwise structurally distinct.
