Product Specific Information
The MIH4 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. PD-1 is expressed predominantly on activated T and B lymphocytes.
Two ligands of PD-1, PD-L1 (B7-H1) and PD-L2 (B7-DC), have overlapping functions and are expressed on normal tissues and activated antigen-presenting cells. The MIH4 antibody recognizes a different epitope than clone J105.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using stimulated normal human peripheral blood cells.
Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
eFluor™ 450 is a replacement for Pacific Blue™, excited at 405 nm and emitting at 445 nm. Ensure the instrument can detect this fluorochrome.

Target Information
T-cell activation is regulated by inhibitory receptors such as PD-1 (PDCD-1), which binds to ligands PD-L1 and PD-L2 on antigen-presenting cells. This binding inhibits immune activation in the absence of danger signals. PD-1-deficient mice exhibit hyperactivated immune systems and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
