Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by several aggregation-prone proteins and maintains partially unfolded proteins for refolding by other chaperones such as HSPA8/HSC70. It does not refold proteins by itself but facilitates degradation of client proteins through lysosomal or proteasomal pathways after receptor-mediated internalization.
Secreted isoform 1 protects cells from apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ligase complexes, promoting proteasomal degradation of target proteins such as COMMD1 and IKBKB, thereby modulating NF-κB transcriptional activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis. Clusterin plays a role in the regulation of cell proliferation.
