Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by various proteins and maintains partially unfolded proteins for refolding by other chaperones. It does not require ATP and does not refold proteins by itself. Binding to cell surface receptors leads to internalization and degradation of the chaperone-client complex. Secreted isoform 1 protects cells from apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ubiquitin ligase complexes to promote proteasomal degradation of target proteins, modulating NF-kappa-B activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis, contributing to regulation of cell proliferation.
