Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Immunoglobulin classes share a basic structure of four polypeptide chains: two heavy chains (five types) and two light chains (kappa and lambda, both ~22.5 kDa).
Kappa and lambda chains each have variable and constant regions, which can be differentiated by the antigenic properties of the constant region.
The ratio of kappa to lambda is approximately 70:30.
Antibodies to the kappa light chain are useful in identifying leukemias, plasmacytomas, and certain non-Hodgkin’s lymphomas.
Demonstration of clonality in lymphoid infiltrates indicates malignancy.
