Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor belonging to the immunoglobulin superfamily.
CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, and a subset of normal and malignant B cells.
Cross-linking CD7 modulates T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation.
CD7 associates directly with phosphoinositol 3'-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic CD34(+) CD38(-) cord blood cell population expressing CD45RA, HLA-DR, and high CD7 levels supports lymphopoiesis, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells.
CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells.
Additionally, CD7 may cooperate with CD28 in Treg function; deficiency in both reduces Treg numbers and suppressive activity.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry of human peripheral blood cells at 5 µL (0.8 µg)/test in 100 µL final volume.
Cell count per test: 10⁵–10⁸ cells (empirically determined).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal NovaFluor staining. Add 5 µL per sample before mixing with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD7 (gp40, Leu9) is a 40 kDa member of the immunoglobulin gene superfamily.
The N-terminal (aa 1–107) is homologous to Ig kappa light chains; the C-terminal domain is proline-rich and forms a stalk structure.
CD7 is expressed on immature/mature T lymphocytes, T cell leukemia, NK cells, and some B cells.
Cross-linking CD7 modulates T and NK cell activity through calcium flux, adhesion, cytokine secretion, and proliferation.
CD7 directly associates with PI3-kinase, inducing D-3 phosphoinositides and tyrosine phosphorylation.
Expression of CD7 serves as a key marker in leukemia diagnostics.
