Product Specific Information
The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes via CD14.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use at 5 µL (0.6 µg) per test in 100 µL total volume. Suitable for 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
- Add 5 µL of CellBlox Plus Blocking Buffer per sample prior to combining with cells.

Target Information
CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a receptor for lipopolysaccharide (LPS) in complex with LBP, mediating immune recognition of bacterial components. Soluble CD14 (sCD14) participates in neutralization of LPS and modulates inflammatory and infectious processes. Alternative splicing results in multiple transcript variants encoding the same isoform.
