Product Specific Information
The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor and member of the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chain, while the C-terminal extracellular domain is proline-rich, forming a stalk projecting the Ig domain.
CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, a subset of B cells, and malignant B cells. Cross-linking CD7 modulates T and NK cell activity by affecting calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates directly with phosphoinositol 3′-kinase, and ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.
A subpopulation of CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels can generate B cells, NK cells, and dendritic cells, but not myeloid or erythroid cells—marking primitive human lymphoid progenitors.
CD7 may cooperate with CD28 in Treg function, as CD28/CD7-deficient mice exhibit reduced Treg numbers and suppressive activity.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.8 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
- Add 5 µL per sample before combining with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor Technology:
Based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized fluorescence signatures minimizing spillover and spread.
Excitation: 552 nm
Emission: 700 nm
Laser: 561 nm (Yellow)
Target Information
CD7 (gp40, Leu9) is a 40 kDa immunoglobulin superfamily member. The N-terminal domain (aa1–107) resembles Ig kappa light chains, while the C-terminal extracellular region is proline-rich, forming a stalk-like structure. CD7 is expressed on immature/mature T cells, T cell leukemias, NK cells, and subsets of normal and malignant B cells. Cross-linking CD7 modulates T and NK cell activation, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. It associates with PI3K, leading to D-3 phosphoinositide production and tyrosine phosphorylation. CD7 is a key diagnostic marker in leukemia research.
