Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor and member of the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chain, while the C-terminal extracellular domain is proline-rich, forming a stalk projecting the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, a subset of B cells, and malignant B cells. Cross-linking CD7 modulates T and NK cell activity by affecting calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates directly with phosphoinositol 3′-kinase, and ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.

A subpopulation of CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels can generate B cells, NK cells, and dendritic cells, but not myeloid or erythroid cells—marking primitive human lymphoid progenitors.

CD7 may cooperate with CD28 in Treg function, as CD28/CD7-deficient mice exhibit reduced Treg numbers and suppressive activity.

Each product contains:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.8 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
